目的:观察左、右归丸对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)大鼠脑组织中凋亡蛋白Fas、Bax表达的影响。方法:用髓鞘碱性蛋白(myelin basic protein68-86,MBP68-86)免疫Lewis大鼠建立EAE模型。观察各组大鼠体重体温变化,饮食和饮水变化,神经功能评分,发病率、死亡率、潜伏期及病程变化。取脑和脊髓,HE染色后进行病理观察;用免疫组化法检测大鼠脑组织中Fas、Bax的表达情况。结果:左归丸组和右归丸组明显减轻病灶区域的炎性细胞浸润,对Fas、Bax的表达均有一定的抑制作用,与激素组类似。结论:左、右归丸均有防治小鼠EAE的作用,其作用机制可能与调节细胞凋亡蛋白Fas、Bax的表达有关。
Objective: To observe the effects of Zuogui Yougui on the expression of Fas and Bax proteins in the brain of experimental autoimmune encephalomyelitis (EAE) rats. Methods: Lewis rats were immunized with myelin basic protein 68-86 (MBP68-86) to establish EAE model. Body temperature changes in body weight, changes in diet and drinking water, neurological function scores, morbidity, mortality, latency and course of disease were observed in each group. The brain and spinal cord were taken out and stained with hematoxylin and eosin for pathological observation. The expression of Fas and Bax in rat brain tissue was detected by immunohistochemistry. Results: Zuoguiwan and Yougui Wan significantly reduce the infiltration of inflammatory cells in the lesion area, and have a certain inhibitory effect on the expression of Fas and Bax, similar to the hormone group. Conclusion: Both Zuogui Pill and Yougui Guiyue can prevent and cure EAE in mice. The mechanism may be related to the regulation of apoptosis proteins Fas and Bax.
