目的观察红景天苷对亚氨基二丙腈诱导的大鼠头部抽动Tourette综合征模型的纹状体多巴胺(DA)水平及抽动行为的影响。方法 Wistar雄性大鼠(40只)随机分为空白组、模型组、氟哌啶醇组和红景天苷组,每组10只。除空白组外均分别以亚氨基二丙腈诱导大鼠头部抽动建立Tourette综合征模型,评估各组给药前后大鼠的刻板行为评分,以及应用高效液相色谱法测定的血浆及纹状体中DA水平变化。结果与模型组相比,给药7 d后红景天苷组及氟哌啶醇组大鼠头部抽动行为明显减少(P<0.01),且血浆及纹状体DA水平明显增高(P<0.01)。结论红景天苷能降低多巴胺能神经系统功能,减少Tourette综合征模型大鼠的抽动行为。
Objective To observe the effect of salidroside on dopamine (DA) levels and twitch behavior in striatum induced by iminodipropionitrile in Tourette’s syndrome of head-twitching rats. Methods Wistar male rats (40) were randomly divided into blank group, model group, haloperidol group and salidroside group, with 10 rats in each group. Except for the blank group, the model of Tourette’s syndrome was induced by iminodipropionitrile-induced head twitch in rats, and the stereotypical scores of rats in each group before and after administration were also evaluated. The plasma and striatum were measured by high performance liquid chromatography DA levels in the body changes. Results Compared with model group, head twitch behavior was significantly decreased in salidroside and haloperidol groups (P <0.01), and DA in plasma and striatum were significantly increased (P < 0.01). Conclusions Salidroside can reduce the function of dopaminergic nervous system and reduce the twitching behavior in rats with Tourette’s syndrome.
