目的:研究膀胱移行细胞癌组织中脆性组氨酸三联体(fragile histidine triad,FH1T)mRNA及蛋白在膀胱移行细胞癌发生、发展中的作用。方法:采用RT-PCR和免疫组化SP法检测FHIT mRNA及蛋白在62例膀胱移行细胞癌组织及35例正常组织中的表达情况。结果:FHIT mRNA在癌组织及正常对照组中的缺失率分别为54.84%和2.86%。随着临床分期,FHIT mRNA和蛋白异常表达呈增高趋势;并且随着膀胱移行细胞癌分化程度呈下降趋势。结论:FHIT基因与膀胱移行细胞癌的发生、发展过程密切相关,FHIT基因的检测可能成为膀胱移行细胞癌的预测指标。
Objective: To study the role of fragile histidine triad (FH1T) mRNA and protein in the occurrence and development of bladder transitional cell carcinoma (BTCC). Methods: The expression of FHIT mRNA and protein in 62 cases of bladder transitional cell carcinoma and 35 cases of normal tissues were detected by RT-PCR and immunohistochemistry. Results: The deletion rates of FHIT mRNA in cancer tissues and normal controls were 54.84% and 2.86%, respectively. With the clinical stage, FHIT mRNA and protein abnormal expression showed an increasing trend; and with the degree of transitional cell carcinoma of bladder showed a downward trend. Conclusion: FHIT gene is closely related to the occurrence and development of bladder transitional cell carcinoma. The detection of FHIT gene may be a predictor of bladder transitional cell carcinoma.
