目的探讨抗脑衰胶囊对人脐带间充质干细胞(h UMSCs)向神经元样细胞分化的影响。方法通过贴壁法分离、培养、扩增脐带间充质干细胞,应用流式细胞仪检测其表面抗原表达。制备抗脑衰胶囊含药血清,诱导h UMSCs,在倒置显微镜下观察细胞形态变化,应用免疫细胞化学方法观察诱导后神经细胞相关基因巢蛋白(Nestin),神经元特异性烯醇化酶(NSE)的表达,并应用RT-PCR技术检测NSE表达。结果抗脑衰胶囊含药血清培养h UMSCs后,Nestin、NSE表达阳性,RT-PCR检测结果显示,诱导后细胞表达NSE。结论抗脑衰胶囊能促进h UMSCs向神经元样细胞分化,是较为理想的诱导剂。
Objective To investigate the effect of Kangnao Capsule on differentiation of human umbilical cord mesenchymal stem cells (h UMSCs) into neuron-like cells. Methods Umbilical cord mesenchymal stem cells were isolated, cultured and expanded by adherence method. The expression of surface antigens was detected by flow cytometry. The serum containing Kangnai Capsule was induced to induce hMSCs, and the morphological changes of cells were observed under an inverted microscope. Nestin, neuron specific enolase (NSE) The expression of NSE was detected by RT-PCR. Results Anti-Nao Fa capsule containing serum h UMSCs cultured after Nestin, NSE expression was positive, RT-PCR results showed that after induction of cells express NSE. Conclusion Kangnai Capsule can promote the differentiation of h UMSCs into neuron-like cells, which is an ideal inducer.
