目的 观察在Wistar大鼠动脉粥样硬化斑块形成过程中NLRP3及IL-1β的mRNA及蛋白质水平表达变化,了解NLRP3/IL-1β信号通路在动脉粥样硬化形成过程中发挥的作用。方法 采取维生素D3联合高脂饲料方法建立动脉粥样硬化模型。并分时间段取材,用实时荧光定量PCR及蛋白免疫印迹法检测NLRP3及IL-1β的mRNA及蛋白水平的表达变化。结果 在Wistar大鼠动脉粥样硬化形成过程中,随着时间的延长,NLRP3及IL-1β的RNA和(或)蛋白表达量则逐渐上升。结论 该研究提示NLRP3/IL-1β在大鼠动脉粥样硬化发展过程中明显增强,其作用和意义有待研究。
Objective To observe the mRNA and protein expression of NLRP3 and IL-1β during the development of atherosclerotic plaque in Wistar rats and to find out the role of NLRP3 / IL-1β signaling in the development of atherosclerosis. Methods Atherosclerosis model was established by vitamin D3 combined with high fat diet. The mRNA and protein levels of NLRP3 and IL-1β were detected by real-time fluorescence quantitative PCR and Western blotting. Results During the development of atherosclerosis in Wistar rats, the expression of RNA and / or protein of NLRP3 and IL-1β gradually increased with time. Conclusions This study suggests that NLRP3 / IL-1β may be significantly increased during the development of atherosclerosis in rats. The role and significance of NLRP3 / IL-1β in the development of atherosclerosis in rats need to be studied.
