本组既往研究发现,针刺备用根猫脊髓背角组织提取液大于50KD组份内存在促神经突起生长的神经营养活性物质。本实验采用双侧备用根猫模型(切除双侧L1~L5,L7~S2背根节,保留L6为备用根),每日针刺动物一侧L6脊神经支配范围的足三里与悬钟、伏兔与三明交两组穴位之一,以进一步证实针刺时该类神经营养活性物质的影响。术后5天取双侧T12~SZ脊髓背角组织,匀浆、离心后,取上清液进行电泳分析,并与鸡胚背根节(DRG)联合培养。结果显示:1.对聚丙烯酸胺梯度电泳的电泳谱分析,发现在相对迁移率(RM)0.11带的蛋白质,针刺侧吸收峰面积百分比(11.78±1.08%)较非针刺侧(10.12f0.83%)明显增高(P<0.05)。2.取RM0.11带及空白凝胶小片分别与DRG联合培养,48小时后测量DRG神经突起平均长度。发现针刺侧(22043±6.01pm)、非针刺侧(187.08±8.01pm)、以及参照组(15577±2.04pm)之间均存在明显差异。以上结果提示,部分去传入猫背角组织中存在某种促神经突起生长的神经营养活性物质,针刺通过增加该活性物质的含量来加强这种促神经突起生长效应。由此推测,部分去传入和针刺引起的这类神经营养活性物质的含量增加,可能与备用根猫在体的侧支出芽有关。
This group of previous studies found that acupuncture spare root cat spinal cord dorsal horn tissue extract greater than 50KD components in the presence of neurite outgrowth neurotrophic substances. In this experiment, bilateral alternate root cat model (excised bilateral L1 ~ L5, L7 ~ S2 dorsal root ganglia, reserved L6 as the backup root), the daily acupuncture animal L6 spinal nerve innervation Zusanli and hanging bell, volts Rabbit and Sanming handed one of two points to further confirm the acupuncture effects of such neurotrophic substances. Twenty-five days after operation, T12-SZ spinal dorsal horn tissues were harvested, homogenized and centrifuged. Supernatants were harvested for electrophoresis analysis and co-cultured with chick embryo dorsal root ganglia (DRG). The result shows: 1. Electrophoretic analysis of polyacrylamide gradient electrophoresis showed that the percent absorption peak area (11.78 ± 1.08%) on the acupuncture side was significantly lower than that on the non-acupuncture side (10 .12f0.83%) was significantly higher (P <0.05). 2. Take RM0.11 strip and blank gels were DRG co-culture, measured 48 hours after the average length of DRG neurites. Significant differences were found between the acupuncture side (22043 ± 6.01pm), the non-acupuncture side (187.08 ± 8.01pm), and the reference group (15577 ± 2.04pm). The above results suggest that some neurotrophic substances which promote the growth of neurites in part of the cat’s dorsal horn tissues are acupunctured to increase the growth promoting effect of the neurite outgrowth by increasing the content of the active substance. It is speculated that some of the detritus and acupuncture caused by the increase in the content of such neurotrophic substances, may be related to spare root cats in the body side buds related.
