目的:研究CXCR4在低氧诱导的上颌窦癌细胞IMC3中的表达及其意义。方法:Real-Time PCR测定低氧6h、12h、24h、48hIMC3中CXCR4的mRNA表达量,并行免疫组织化学研究。结果:CXCR4mRNA正常状态下为0.035,低氧6h、12h、24h、48h分别为0.283、0.313、0.426、0.510。除低氧6h和12h组相比差异无统计学意义外,其余各组间差异有统计学意义(P<0.05)。免疫组织化学结果表明,低氧下CXCR4的mRNA表达主要在细胞膜和细胞质,少量在细胞核。结论:低氧能诱导IMC3细胞CXCR4mRNA的表达,随低氧时间延长而增高,有明显时间性。
Objective: To investigate the expression of CXCR4 in hypoxia-induced maxillary sinus cancer cells IMC3 and its significance. Methods: Real-time PCR was used to detect the mRNA expression of CXCR4 in 6 hours, 12 hours, 24 hours and 48 hours hypoxia, and immunohistochemistry was performed. Results: The normal CXCR4 mRNA was 0.035, hypoxia 6h, 12h, 24h, 48h were 0.283,0.313,0.426,0.510 respectively. Except for hypoxia, there was no significant difference between 6h and 12h groups (P <0.05). Immunohistochemical results showed that under hypoxia CXCR4 mRNA expression mainly in the cell membrane and cytoplasm, a small amount in the nucleus. Conclusion: The hypoxia can induce the expression of CXCR4 mRNA in IMC3 cells, which increases with the prolongation of hypoxia time.
