目的:观察通过饮食劳倦所伤复合腹腔注射丁胺卡那霉素及链脲佐菌素作为造模要素能否成功复制出脾肾气虚证2型糖尿病病证结合动物模型。方法:健康雄性SD大鼠60只,体质量(180±20)g,按体质量随机分成二组:空白对照组(n=20)和模型组(n=40)。模型组大鼠单日用200%的大黄浓煎液灌胃,4 m L/只,游泳至耐力极限;双日高糖高脂膳食持续喂养,共计12周,第4周后给予丁胺卡那霉素及链脲佐菌素腹腔注射。结果:模型组大鼠造模4周后出现游泳耐力逐日下降,体毛光亮度减退,倦怠,懒动,喜聚堆、弓背蜷缩,粪便时软、时塘,血糖增高;第6周后体质量开始显著增加;甘油三酯和高密度脂蛋白分别从第2周后表现出异常;实验结束后模型组出现高胰岛素血症、胰岛素抵抗,符合本研究拟建立的脾肾气虚证、2型糖尿病的评估标准。结论:通过饮食劳倦所伤复合腹腔注射丁胺卡那霉素及链脲佐菌素造模方法可成功复制出脾肾气虚证2型糖尿病病证结合动物模型。
OBJECTIVE: To observe whether the combination of amikacin and streptozotocin (STZ) induced by diet-induced fatigue could successfully replicate the combined animal model of type 2 diabetes with syndrome of spleen and kidney-qi deficiency. Methods: Sixty male Sprague Dawley rats weighing 180 ± 20 g were randomly divided into two groups according to body weight: blank control group (n = 20) and model group (n = 40). Rats in the model group were orally gavaged with 200% rhubarb concentrate on a daily basis, swimming to endurance limit of 4 m L / bird; high-fat and high-fat diet for two days were fed continuously for 12 weeks, Natamycin and streptozotocin intraperitoneal injection. Results: After 4 weeks of modeling, the endurance of swimming in the model rats decreased day by day, body lightening, fatigue, laziness, hippocampus, curled bow, stool soft, The quality began to increase significantly; triglyceride and high density lipoprotein showed abnormalities after 2 weeks; hyperinsulinemia and insulin resistance appeared in the model group after the experiment, in line with the model of spleen and kidney qi deficiency, type 2 Diabetes evaluation criteria. CONCLUSION: The combination of amlodipine and streptozotocin in rats induced by diet fatigue can replicate successfully the combined animal model of type 2 diabetes with syndrome of spleen and kidney qi deficiency.
