本研究的目的是建立适合于人成纤维细胞干扰素(HuIFN-β)大规模生产的二倍体细胞株。从8株新建的细胞中选择出的一株人胚肌皮细胞——SM_(15)的特点如下:(1) 生长速率快,细胞1∶2接种形成单层的时间为24h,维持时间多于15天。(2) 总群体倍增水平为57.7。(3) 通过起动和NDV超诱导,干扰素的平均产量为23000IU/ml。(4) 冻存对该株细胞的干扰素产量无影响。(5) SM_(15)的核型为正常人二倍体核型,即46XX,可用于临床人β干扰素的大规模生产。
The purpose of this study was to establish a diploid cell line suitable for large-scale production of human fibroblast interferon (HuIFN-β). The characteristics of SM_ (15), a human embryonic myofibroblast selected from 8 newly-established cells, are as follows: (1) The growth rate is fast, the time of inoculating 1: 2 cells into a monolayer is 24 hours, and the maintenance time At 15 days. (2) The total population doubling level was 57.7. (3) The average yield of interferon was 23000 IU / ml by induction and NDV superinduction.
