目的探讨组蛋白去乙酰化酶1(HDAC1)水平改变在二苯乙烯苷(TSG)对沙鼠脑缺血/再灌注(I/R)损伤中的作用。方法 64只沙鼠随机分为假手术组、脑I/R模型对照组、TSG处理组和依达拉奉对照组。Morris水迷宫实验观察TSG对脑I/R鼠学习记忆功能的影响;神经功能评分、氯化三苯基四氮唑(TTC)染色观察TSG对经I/R脑组织的保护作用;免疫组织化学SP法观察脑组织中HDAC1蛋白的表达。Western blotting和RT-PCR检测HDAC1蛋白和mRNA的水平。结果与模型组比较,TSG对I/R鼠的脑组织有保护作用,能够改善模型鼠学习记忆能力,使模型鼠脑组织HDAC1蛋白和mRNA水平明显升高。结论 TSG能够改善脑I/R损伤模型鼠认知功能,其作用机制与组蛋白乙酰化和去乙酰化动态平衡有关。
Objective To investigate the role of histone deacetylase 1 (HDAC1) levels in the injury of gerbil brain ischemia / reperfusion (I / R) by stilbene glycoside (TSG). Methods 64 gerbils were randomly divided into sham operation group, brain I / R model control group, TSG treatment group and edaravone control group. Morris water maze test was used to observe the effect of TSG on learning and memory in I / R rats. Neurological function score, TTC staining were used to observe the protective effect of TSG on I / R brain tissue. Immunohistochemistry SP method to observe the expression of HDAC1 protein in brain tissue. The levels of HDAC1 protein and mRNA were detected by Western blotting and RT-PCR. Results Compared with the model group, TSG could protect the brain tissue of I / R mice and improve the ability of learning and memory in model mice, and significantly increase the level of HDAC1 protein and mRNA in brain of model mice. Conclusion TSG can improve the cognitive function in the rat model of cerebral I / R injury, and its mechanism is related to the dynamic balance of histone acetylation and deacetylation.
