目的:探讨胎盘部位脂联素及其受体基因mRNA表达在妊娠期糖尿病病理状态下的变化趋势及与新生儿体重、新生儿Ponderal指数(PI)之间的关系。方法:对26例妊娠期糖尿病产妇、30例正常产妇采用逆转录聚合酶链反应(RT-PCR)技术检测胎盘脂联素及其受体基因mRNA的表达,并对其表达水平进行半定量分析。结果:①脂联素及其受体1/2基因mRNA表达于胎盘组织;②妊娠期糖尿病组胎盘脂联素基因mRNA表达水平(0.75±0.19)显著低于正常对照组(1.17±0.17),差异有统计学意义(P<0.05);妊娠期糖尿病组胎盘AdipoR1基因mRNA的表达水平(2.08±0.67)与正常对照组(1.98±0.68)比较,差异无统计学意义(P>0.05);妊娠期糖尿病组胎盘AdipoR2基因mRNA的表达水平(1.58±0.36)显著高于正常对照组(1.19±0.25),差异有统计学意义(P<0.05);③胎盘脂联素基因mRNA表达水平与新生儿出生体重、PI呈显著负相关关系;胎盘AdipoR2基因mRNA表达水平与新生儿出生体重、PI无相关性。结论:①胎盘组织中存在脂联素及其受体基因的表达;②胎盘来源的脂联素及其受体可能参与了胎儿生长发育的调节。
OBJECTIVE: To investigate the changes of adiponectin and its receptor gene mRNA expression in placental tissue during the gestational diabetes mellitus (DM) and its relationship with body weight and Ponderal index (PI) in neonates. Methods: Twenty-six pregnant women with gestational diabetes mellitus and 30 normal pregnant women were tested for mRNA expression of adiponectin and its receptor by reverse transcription-polymerase chain reaction (RT-PCR), and their expression levels were semi-quantitatively analyzed . Results: (1) Adiponectin and its receptor 1/2 gene mRNA expression in placenta tissue; (2) Placental adiponectin gene mRNA expression level (0.75 ± 0.19) in gestational diabetes mellitus group was significantly lower than that in the normal control group (1.17 ± 0.17) (P <0.05). There was no significant difference in the expression of placental AdipoR1 gene mRNA in gestational diabetes mellitus (2.08 ± 0.67) compared with that in normal control group (1.98 ± 0.68) (P> 0.05), and the difference was statistically significant The placental AdipoR2 gene mRNA expression level (1.58 ± 0.36) in stage diabetes mellitus group was significantly higher than that in control group (1.19 ± 0.25), the difference was statistically significant (P <0.05); ③In placental adiponectin gene mRNA expression level Birth weight and PI had a significant negative correlation. The placental AdipoR2 mRNA expression level had no correlation with the newborn’s birth weight and PI. Conclusion: ① The expression of adiponectin and its receptor gene exists in placenta tissue; ② Placenta adiponectin and its receptor may be involved in the regulation of fetal growth and development.
