采用体外造血细胞培养技术,应用高温、利多卡因、足叶乙苷(足叶乙甙,VP-16)联合法,研究其在体外对白血病细胞的杀伤作用。结果表明:高温、利多卡因、VP-16三者联用比高温+利多卡因、高温+VP-16,能更有效地杀伤白血病细胞,而正常粒单系克隆形成单位(GM-CFU)仍约有50%存活。41℃60min+0.75mmol/L利多卡因+8.5μmol/LVP-16联合处理后,HL-60、U937、K562细胞分别只存活0.01%、9.40%、33.66%。另外,从这个联合方案说明对于不同类型的白血病效果不一,在其自身骨髓移植(ABMT)时,体外净化骨髓要采用不同的净化方案。
In vitro hematopoietic cell culture technique was used to study the cytotoxicity against leukemia cells in vitro by high temperature, lidocaine and etoposide (VP-16). The results showed that the combination of high temperature, lidocaine and VP-16 could kill leukemia cells more efficiently than the combination of high temperature plus lidocaine and high temperature + VP-16. However, the expression of GM-CFU About 50% survive. The survival rates of HL-60, U937 and K562 cells were only 0.01%, 9.40% and 33.66% when the cells were treated with 41mmol / L 0.75mmol / L lidocaine + 8.5μmol / LVP-16 for 41min. In addition, the synergistic effect of different combinations of leukemias is demonstrated by this combination protocol, with different decontamination protocols being used to decontaminate bone marrow in vitro during its own bone marrow transplantation (ABMT).
