目的:研究家兔补肾化瘀方含药血清对宫腔粘连子宫内膜基质细胞的生长周期及MMP-9因子的影响,探讨其部分作用机制。方法:选取宫腔粘连患者子宫内膜组织,制备宫腔粘连子宫内膜基质细胞模型。将家兔分为中药高、中、低剂量组及西药补佳乐组、空白组5组,用不同浓度补肾化瘀中药以及西药补佳乐灌胃7d,心脏采血并取含药血清,用家兔含药血清干预细胞模型,应用流式细胞检测仪检测细胞周期,ELISA法检测MMP-9因子的表达。结果:各组家兔含药血清作用于宫腔粘连患者子宫内膜基质细胞模型24h后,与空白组比较,各给药组的增殖指数(PI)和MMP-9含量均增高,凋亡指数(API)均降低,S期细胞百分比均增高,差异有统计学意义(P<0.01)。与西药组比较,中药各组PI增高,API降低,S期细胞百分比增高,中药中、低剂量组MMP-9表达减少,差异有统计学意义(P<0.01);但中药高剂量组MMP-9表达与西药组差异无统计学意义(P>0.05)。与中药中剂量组比较,中药高剂量组MMP-9表达增多,PI增高,API降低,S期细胞百分比增高,中药低剂量组PI降低,API增高,差异均有统计学意义(P<0.01);中药中、低剂量组的S期细胞百分比差异无统计学意义(P>0.05)。与中药低剂量组比较,中药高、中剂量组与西药组MMP-9表达增多,差异均具有统计学意义(P<0.01)。结论:含有补肾化瘀方的家兔含药血清可以提高宫腔粘连子宫内膜基质细胞中MMP-9因子的水平,促进宫腔粘连子宫内膜基质细胞的增殖,并降低其凋亡,不同浓度的补肾化瘀方对于宫腔粘连子宫内膜基质细胞的影响也有区别。
Objective: To study the effect of BuShun HuayuFang decoction serum on the growth cycle and MMP-9 of uterine cavity adhesion endometrial stromal cells and to explore its partial mechanism. Methods: Endometrial tissue from patients with intrauterine adhesions was selected to prepare intrauterine adhesion endometrial stromal cells model. The rabbits were divided into high, medium and low dose groups of traditional Chinese medicine and western medicine nociceptive group, blank group of 5 groups, with different concentrations of kidney and blood stasis medicine and western medicine nourishing music stomach 7d, Rabbit drug-containing serum intervention cell model, flow cytometry was used to detect cell cycle, ELISA assay MMP-9 factor expression. Results: After treated with serum of rabbits in each group for 24 h, the proliferative index (PI) and MMP-9 content in each group were higher than those in the blank group, and the apoptosis index (API) were decreased, the percentage of S phase cells were increased, the difference was statistically significant (P <0.01). Compared with the western medicine group, the PI in each group increased, the API decreased, and the percentage of S phase cells increased. The expression of MMP-9 in middle and low dose group decreased significantly (P <0.01) 9 expression and western medicine group had no significant difference (P> 0.05). Compared with the traditional Chinese medicine medium dose group, the expression of MMP-9 increased, the PI increased, the API decreased, the percentage of S phase cells increased, the PI decreased and the API increased in the traditional Chinese medicine high dose group (P <0.01) There was no significant difference in the percentage of S phase cells between Chinese medicine and low dose groups (P> 0.05). Compared with the traditional Chinese medicine low-dose group, the expression of MMP-9 in high-dose, middle-dose and western medicine groups increased, the difference was statistically significant (P <0.01). CONCLUSION: The serum of rabbits containing Bushen Huayu Recipe can increase the level of MMP-9 in intrauterine adhesion endometrial stromal cells, promote the proliferation of uterine cavity adhesion endometrial stromal cells and reduce their apoptosis, Concentration of Kidney Huayu side of the uterine cavity adhesion endometrial stromal cells are also different.
