With the young leaves of L. spicata as the materials, conditions for the callus induction and differentiation, the adventitious bud differentiation and rooting, and the transplanting and field planting of tube seedlings were studied. Clone technology for the young leaves of L. spicata was established. Results showed that MS + 0.4 mg/L 6-BA + 1.0 mg/L NAA + 1.5 mg/L 2,4-D was the ideal culture medium for callus induction from young leaves of L. spicata; 1/2 MS + 1.2 mg/L AgNO3 + 0.5 mg/L BA + 0.1 mg/L NAA was the ideal culture medium for the callus and adventitious bud differentiation; and 1/2 MS+0.2 mg/L IAA +0.1 mg/L NAA was the ideal culture medium for the rooting of adventitious buds of L. spicata. Tube seedlings of L. spicata had the characteristics of vigorous growth status and developed root system, while other biological characteristics of tube seedlings stayed the same.
