目的研究前胡丙素对大鼠缺血-再灌注(I-R)心肌肌浆网钙泵(SERCA)的影响及机制。方法 24只雄性SD大鼠随机均分为正常对照组、I-R组、前胡丙素+I-R组和聚乙二醇400+I-R组。检测再灌注15min时冠状动脉流出量(CF)和冠状动脉流出液中乳酸脱氢酶(LDH)漏出量,采用改良对硝基苯酚棕榈酸酯(p-NPP)法和Millipore过滤技术测定SERCA的活性和三磷酸腺苷依赖的SERCA45 Ca2+摄取率,Western blot法检测心肌细胞SERCA蛋白表达水平。结果与I-R组相比,前胡丙素+I-R组CF、SERCA蛋白表达量、SERCA的活性和45 Ca2+摄取率增高(P<0.01),LDH漏出量降低(P<0.01)。结论前胡丙素对离体大鼠心脏I-R损伤有保护作用,可能与其减轻I-R损伤引起的心肌SERCA功能和蛋白表达抑制有关。
Objective To investigate the effect and mechanism of PRA on SERCA in rats with myocardial ischemia reperfusion (I-R). Methods Twenty-four male SD rats were randomly divided into normal control group, I-R group, pre-flavanin + I-R group and polyethylene glycol 400 + I-R group. Coronary artery outflow (CF) and lactate dehydrogenase (LDH) leakage in the coronary effusion were measured at 15 min after reperfusion, and the levels of SERCA were measured by modified p-nitrophenol palmitate (p-NPP) and Millipore filtration Activity and adenosine triphosphate-dependent SERCA45 Ca2 uptake rate, Western blot detection of myocardial cell SERCA protein expression levels. Results Compared with I-R group, the levels of CF, SERCA protein, SERCA and 45 Ca 2+ uptake in pre-Bupropion + I-R group were significantly increased (P <0.01) and LDH leakage was decreased (P <0.01). Conclusions Preuplularin has a protective effect on I-R injury in isolated rat hearts, which may be related to the decrease of SERCA function and protein expression induced by I-R injury.
