目的使培养肝细胞在体外重建细胞极性。方法利用三明治构型培养成鼠肝细胞并观察其形态、特异性膜区域蛋白分布并以单层胶原培养肝细胞作对照。结果三明治构型肝细胞呈单层生长 ,形成肝板样结构 ,伴胆小管网络形成 ,免疫组织化学证明肝细胞膜区域蛋白呈特异性分布。而在对照组 ,肝细胞基本没有形成肝板样结构 ,肝细胞膜区域蛋白没有形成特异性分布。结论三明治构型培养肝细胞在体外重建细胞极性 ,保留了体内肝细胞的特点
Objective To cultivate hepatocytes to reconstitute cell polarity in vitro. Methods The sandwich hepatocytes were cultured and the morphology and distribution of proteins in specific membrane regions were observed. Results Sandwich-type hepatocytes grew in monolayer and formed hepatic platelet-like structures with the formation of bile duct network. Immunohistochemistry showed that the protein in liver cell membrane was specifically distributed. However, in the control group, hepatic platelet-like structures were almost not formed in the control group, and no specific distribution was found in the hepatocyte membrane protein. Conclusion Sandwich configuration of cultured hepatocytes reconstructs the cell polarity in vitro and retains the characteristics of hepatocytes in vivo
