目的 :建立青蒿琥酯的紫外分析方法。方法 :探讨了青蒿琥酯水解温度、碱浓度以及水解时间对青蒿琥酯水解产物的影响 ,同时进行紫外测定法的方法学考察。结果 :青蒿琥酯的最佳水解条件为 :83℃ ,0 .1mol/L氢氧化钠 ,水解 1h可得到完全稳定的水解产物 ,并确定 2 3 8nm为紫外最大吸收波长。线性范围 1 3~ 1 95mg/L ,最低检测限为 5mg/L。结论 :青蒿琥酯在碱性条件下水解后进行紫外测定 ,是一稳定可行的分析方法
Objective : To establish a UV analysis method for artesunate. Methods: The effects of artesunate hydrolysis temperature, alkali concentration and hydrolysis time on the hydrolysate of artesunate were investigated. At the same time, the methodological investigation of UV assay was performed. Results : The optimal hydrolysis conditions for artesunate were: 83°C, 0.1 mol/L sodium hydroxide, and 1 h hydrolysis yielded a completely stable hydrolysate, and 238 nm was determined as the maximum UV absorption wavelength. The linear range is from 1 3 to 195 mg/L with a detection limit of 5 mg/L. Conclusion : It is a stable and feasible method for determination of artesunate after UV hydrolysis under alkaline conditions.
