利用旧动物室改造成具有恒温、恒湿、洁净的饲育室,配合无菌操作技术,应用层流架饲养从上海肿瘤所引进的SPF裸鼠,经过四年对动物育种,饲养和无菌操作技术进行探讨。我们成功地培育了300只SPF裸鼠用于人胃高分化腺癌7911侈植瘤、人胃低分化腺癌MGC80-3裸鼠肝转移、人肝癌组织裸鼠移植瘤、人肝癌细胞株BEL-7102移植瘤、3.4苯丙吡诱发裸鼠皮肤癌等模型。在动物育种方面我们采用BALB/c非裸近交产生基因稳定核心群,BALB/c裸鼠与BALB/c核心群回交以保持BALB/c裸鼠基因的稳定性和一致,利用回交产生的雌性半裸鼠和维性全裸鼠生产裸鼠,淘汰半裸鼠。以此循环能准确断定仔鼠基因型,使留种,配种及繁殖方便准确。此繁殖方法生产断乳的仔鼠比杂交法断乳仔鼠平均体重增加2g,半裸雌鼠受孕率达82%,28和60日龄课鼠与同龄半裸或非裸鼠体重相差无几,已接近国内同行繁育裸鼠水平。在改造旧动物房繁殖裸鼠同时,我们严格采用无菌衣帽穿戴、消毒和无菌操作技术使层流架和房间每平皿菌落低于国外同类饲育室平均标准。
