目的探讨人类转移抑制基因KiSS1及其受体hOT7T175在乳腺癌中的表达情况及临床意义。方法以逆转录聚合酶链反应(RTPCR)方法检测40例癌组织和癌旁正常组织中KiSS1mRNA和hOT7T175mRNA的表达。结果40例乳腺癌组织中,17例(42.5%)KiSS1基因表达阳性,10例(25%)hOT7T175基因表达阳性;正常组织中29例(72.5%)KiSS1基因表达阳性,20例(50%)hOT7T175基因表达阳性。KiSS1基因和hOT7T175基因失表达与临床分期、肿瘤大小等均无显著相关性,而KiSS1基因在淋巴结阴性者表达阳性率为63.6%,阳性者仅为16.7%,两者差异有统计学意义(P<0.01)。结论KiSS1基因失表达与乳腺癌的淋巴结转移密切相关,提示该基因的表达可能是乳腺癌中一个重要的监控淋巴结转移的生物学标志。
Objective To investigate the expression of human metastasis suppressor gene KiSS1 and its receptor hOT7T175 in breast cancer and its clinical significance. Methods The expressions of KiSS1 mRNA and hOT7T175 mRNA in 40 cases of cancerous tissues and adjacent normal tissues were detected by reverse transcriptase polymerase chain reaction (RTPCR). Results KiSS1 gene expression was positive in 17 cases (42.5%) and hOT7T175 gene in 10 cases (25%). The positive expression rate of KiSS1 gene in 29 cases (72.5%) was normal in 20 cases (50% hOT7T175 gene expression was positive. KiSS1 gene and hOT7T175 gene loss of expression and clinical stage, tumor size were no significant correlation, while KiSS1 gene expression in lymph node-negative was 63.6%, positive was only 16.7%, the difference was statistically significant (P <0.01). Conclusion The loss of KiSS1 gene expression is closely related to lymph node metastasis in breast cancer, suggesting that the expression of this gene may be an important biological marker for monitoring lymph node metastasis in breast cancer.
