目的研究通心络对糖尿病大鼠心肌组织糖基化终末产物受体(RAGE)及核转录因子(NF-κB)表达的影响,探讨其对糖尿病大鼠心肌的保护作用及机制。方法雄性W istar大鼠60只,随机选出15只作为正常对照组(C组,n=15),剩余45只尾静脉注射0.1%链脲佐菌素(STZ)建立糖尿病模型,成模大鼠40只,随机分为糖尿病模型组(DM组,n=20)和糖尿病通心络治疗组(TXL组,n=20),通心络500m g/(kg.d)。喂养24周后,各组大鼠存活数为C组12只、DM组9只、TXL组13只,处死大鼠迅速心脏穿刺取血,测空腹血糖、糖化血红蛋白(HbA1c)及糖基化终末产物(AGEs)等指标。RT-PCR及W estern b lot法测定大鼠心肌组织中RAGE和NF-κB的表达。结果与C组相比,DM组大鼠血清中AGEs水平及心肌组织中RAGE和NF-κB的表达显著升高(P均<0.05)。经通心络治疗后,TXL组大鼠血清中AGEs的水平与DM组无统计学差异(P>0.05),但心肌组织中RAGE和NF-κB的表达较DM组显著降低(P均<0.05)。结论通心络可能通过抑制糖尿病大鼠心肌组织RAGE及NF-κB的表达而对糖尿病大鼠心肌组织起到一定的保护作用。
Objective To investigate the effect of Tongxinluo capsule on the expression of RAGE and NF-κB in diabetic rats, and to explore its protective effect on diabetic rats and its mechanism. Methods Fifty-six male Wistar rats were randomly divided into normal control group (group C, n = 15) and the remaining 45 rats were injected with 0.1% streptozotocin (STZ) to establish a diabetic model. Forty mice were randomly divided into diabetic model group (DM group, n = 20) and diabetic Tongxinluo treatment group (TXL group, n = 20) and Tongxinluo 500 m g / (kg · d). After 24 weeks of feeding, the survival rate of rats in each group was 12 in group C, 9 in DM group and 13 in TXL group. The rats were sacrificed by rapid cardiac puncture, and fasting blood glucose, glycosylated hemoglobin (HbA1c) End products (AGEs) and other indicators. RT-PCR and Western blot were used to detect the expression of RAGE and NF-κB in rat myocardium. Results Compared with group C, the levels of AGEs in serum and the expressions of RAGE and NF-κB in myocardium of DM group were significantly increased (all P <0.05). The level of AGEs in TXL group was not significantly different from that in DM group (P> 0.05). However, the expression of RAGE and NF-κB in TXL group was significantly lower than that in DM group (all P <0.05) ). Conclusion Tongxinluo may protect myocardial tissue of diabetic rats by inhibiting the expression of RAGE and NF-κB in diabetic rats.
