将合浦珠母贝三倍体的卵与二倍体的精子授精 ,用 0 .5 μg/mL细胞松弛素B抑制受精卵第一极体的释放诱导四倍体。研究了处理起始时间及持续时间对胚胎孵化率和四倍体诱导率的影响及幼虫的生长及存活。实验结果表明 :持续时间与胚胎孵化率呈负相关 ,而与四倍体诱导率呈正相关 ,持续时间一般为1 5~ 1 8min ,处理起始时间一般在第一极体出现前 3~ 5min。在胚胎期 ,四倍体诱导率平均为 2 0 %。在幼虫培养阶段 ,幼虫死亡严重 ,收苗率极低 ,平均只有 0 .0 2 %。处理组与未处理组的生长差异不明显 (p >0 .0 5 ) ,但两组幼虫生长明显慢于二倍体组 (p <0 .0 5 )。
The oocyte and diploid sperm of Ophiopogon japonicus were inoculated with 0,5 μg / mL cytochalasin B to inhibit the release of the first polar body of the fertilized egg to induce tetraploids. The effects of treatment start time and duration on embryo hatching rate and tetraploid induction rate and larval growth and survival were studied. The results showed that the duration was negatively correlated with the embryo hatching rate, but positively related to the tetraploid induction rate. The duration was generally 15 to 18 min. The initial treatment time was generally 3 to 5 min before the first polar body appeared. In the embryo, tetraploid induction rate was 20% on average. During the larval stage, the larvae died seriously, with very low seedling recovery rates, averaging only 0.022%. There was no significant difference in growth between treatment group and untreated group (p> 0.05), but larval growth of two groups was obviously slower than that of diploid group (p <0.05).
