目的 :为了探讨中性粒细胞 (PMN)的自然凋亡和PMN凋亡对细胞吞噬、趋化和活性氧 (ROS)生成等功能的影响。方法 :采用光镜、电镜、电泳和流式细胞仪等研究手段探讨PMN的自然凋亡 ;通过葡萄球菌法、滤膜法和化学发光的方法研究凋亡PMN吞噬、趋化和ROS生成的改变。结果 :分离健康成人外周血PMN ,在体外进行 10 %血清培养 ,随着培养时间的延长 ,PMN凋亡率升高 ,同时PMN的吞噬、趋化以及ROS生成等功能都不同程度地受损。结论 :外周血PMN在体外培养发生自然凋亡 ,PMN的凋亡与细胞功能相关。
Objective: To investigate the effects of natural apoptosis of neutrophils (PMN) and apoptosis of PMN on the function of phagocytosis, chemotaxis and reactive oxygen species (ROS) generation. Methods: The natural apoptosis of PMN was investigated by light microscope, electron microscope, electrophoresis and flow cytometry. The phagocytosis, chemotaxis and ROS production of apoptotic PMN were studied by staphylococcus aureus, membrane filtration and chemiluminescence . Results: PMN was isolated from peripheral blood of healthy adults and was cultured in vitro in 10% serum. The apoptosis rate of PMN increased with the prolongation of culture time, and the functions of PMN phagocytosis, chemotaxis and ROS production were all impaired to varying degrees. CONCLUSION: Peripheral blood PMN undergoes natural apoptosis in vitro and apoptosis of PMN correlates with cell function.
