目的探讨过量醛固酮对SD大鼠血糖和胰岛素敏感性的影响。方法雄性SD大鼠随机分为正常对照组(NC)、醛固酮组(ALD)、醛固酮+安体舒通组(ALD+SPI)。观察4周,每周测量空腹血糖、体重和血压,实验第4周行腹腔葡萄糖耐量和腹腔胰岛素耐量试验,检测血脂联素水平。结果醛固酮作用4周,SD大鼠空腹血糖变化无统计学意义(P>0.05);ALD组腹腔葡萄糖耐量试验结果与NC组比较差异无统计学意义(P>0.05);腹腔胰岛素耐量试验显示ALD组1 5和30 min血糖下降幅度小于NC组(P<0.05)。ALD组血清脂联素水平较NC组下降22.79%(P<0.05)。结论过量醛固酮作用4周可导致SD大鼠胰岛素抵抗。
Objective To investigate the effects of excessive aldosterone on blood glucose and insulin sensitivity in SD rats. Methods Male SD rats were randomly divided into normal control group (NC), aldosterone group (ALD), aldosterone + spironolactone group (ALD + SPI). After 4 weeks of treatment, fasting blood glucose, body weight and blood pressure were measured weekly. Peritoneal glucose tolerance and peritoneal insulin tolerance test were performed in the fourth week of the experiment to detect the level of adiponectin. Results After 4 weeks of aldosterone treatment, the fasting blood glucose in SD rats had no significant difference (P> 0.05). There was no significant difference between the two groups in ALD glucose tolerance test (P> 0.05). The peritoneal insulin tolerance test showed that ALD The decrease of blood glucose in group 1, 5 and 30 min was less than that in group NC (P <0.05). Serum adiponectin levels in ALD group decreased by 22.79% (P <0.05) compared with NC group. Conclusion Overdose of aldosterone for 4 weeks can lead to insulin resistance in SD rats.
