目的:观察健脾软肝方对四氯化碳大鼠肝纤维化模型中TGF-β/Smad信号转导通路的影响。方法:复制CCl4大鼠肝纤维化模型,观察不同剂量健脾软肝对模型大鼠肝纤维化TGF-β/Smad通路各位点的影响。利用RT-PCR方法检测TGF-β1mRNA的表达水平,免疫组化法检测TGF-β1受体的表达和Western Blot法检测大鼠肝组织中Smad3、Smad7蛋白的表达。结果:健脾软肝方可以下调TGF-β1mRNA和TGF-β1受体的表达,调节Smad3/Smad7的比例。结论:健脾软肝方对TGF-β/Smad细胞信号转导途径中多个位点均有抑制作用。
Objective: To observe the effects of Jianpi Ruangan prescription on TGF-β / Smad signal transduction pathway in carbon tetrachloride-induced hepatic fibrosis model. Methods: The CCl4 rat liver fibrosis model was duplicated and the effect of different doses of Jianpi soft liver on TGF-β / Smad pathway in hepatic fibrosis was observed. The expression of TGF-β1 mRNA was detected by RT-PCR. The expression of TGF-β1 receptor was detected by immunohistochemistry and the expression of Smad3 and Smad7 in rat liver tissue by Western Blot. Results: Jianpi Ruangan Decoction can down-regulate the expression of TGF-β1 mRNA and TGF-β1 receptor and regulate the ratio of Smad3 / Smad7. Conclusion: Jianpi Ruangan Decoction can inhibit multiple sites in signal transduction pathway of TGF-β / Smad cells.
