目的探讨桂皮醛对肾阳虚大鼠γ-氨基丁酸(GABA)受体mRNA表达的影响。方法采用腺嘌呤皮下注射(剂量为0.2 g·kg-1·d-1)构建肾阳虚大鼠模型,桂皮醛灌胃(高、中、低剂量分别为0.4,0.2,0.1 g·kg-1),观察各组大鼠外观及肾脏切片,测定自主活动度,实时荧光定量PCR法测定GABAa受体mRNA的表达。结果造模后,大鼠睾丸增大,体重减轻(P<0.05);肾皮质固缩,与髓质界限不清,有灰褐色沉积物;与模型组比较,各给药组大鼠自主活动度明显降低(P<0.05),GABAa受体mRNA表达显著增高(P<0.05)。结论桂皮醛短时间内未能明显改善阳虚症状,但可能通过提高GABAa受体mRNA表达水平发挥镇静作用。
Objective To investigate the effects of cinnamaldehyde on the mRNA expression of γ-aminobutyric acid (GABA) receptor in kidney yang deficiency rats. Methods A rat model of deficiency of kidney-yang was established by subcutaneous injection of adenine (0.2 g · kg-1 · d-1). Cinnamic aldehyde was administered intragastrically (high, medium and low doses were 0.4,0.2,0.1 g · kg- 1), the appearance of rat and kidney sections were observed, the degree of voluntary activity was measured, and the expression of GABAa receptor mRNA was detected by real-time fluorescence quantitative PCR. Results After the model was established, the testis of rats was enlarged and the body weight was decreased (P <0.05). The renal cortex was constricted and the boundary between the medulla and the medulla was unclear. There was gray-brown sediment. Compared with the model group, (P <0.05), and the expression of GABAa receptor mRNA was significantly increased (P <0.05). Conclusion Cinnamaldehyde can not significantly improve the symptoms of Yang deficiency in a short time, but may play a role in sedation by increasing the expression of GABAa receptor mRNA.
