目的观察四物降脂颗粒对过氧化氢(H_2O_2)致人脐静脉内皮细胞(HUVECs)损伤的保护作用。方法制备四物降脂颗粒大鼠含药血清,随机分为4组:空白组,H_2O_2模型组,药物组,丹参素对照组。倒置显微镜下观察细胞形态、密度;噻唑蓝(MTT)法检测细胞活力;比色法测定细胞培养液中超氧化物歧化酶(SOD)活性及细胞内丙二醛(MDA)含量;免疫细胞化学方法测定TPA,PAI-1含量。结果四物降脂颗粒含药血清可抑制过氧化氢所致的细胞活力降低,减少MDA生成,提高SOD活性;并可升高TPA,PAI-1含量。结论四物降脂颗粒含药血清对过氧化氢所致HUVECs损伤具有明显的保护作用。
Objective To observe the protective effects of Siwu Jiangzhi Granules on the injury of human umbilical vein endothelial cells (HUVECs) induced by hydrogen peroxide (H 2 O 2). Methods Four drug-containing serum of lipid-lowering rats were randomly divided into 4 groups: blank group, H 2 O 2 model group, drug group and Danshensu control group. Cell morphology and density were observed under an inverted microscope. Cell viability was assayed by MTT assay. SOD activity and malondialdehyde (MDA) content in cell culture medium were determined by colorimetric assay. Immunocytochemistry Determination of TPA, PAI-1 content. Results SiLi Jiangzhi Granules could inhibit the decrease of cell viability induced by hydrogen peroxide, decrease the production of MDA, increase the activity of SOD and increase the contents of TPA and PAI-1. Conclusion Siwu Jiangzhi granule contains serum on hydrogen peroxide-induced HUVECs injury has a significant protective effect.
