目的探究扁杏仁肽对人脐静脉内皮细胞(human umbilical vascular endothelial cells,HUVECs)内皮功能的影响。方法体外培养HUVECs,以150,300和600μg·mL~(-1)不同剂量的扁杏仁全肽、扁杏仁谷蛋白肽给药,进行细胞生长增殖试验及细胞培养液中一氧化氮(NO)和内皮素(ET)含量的检测。结果细胞培养48h后,与对照组相比,所有剂量的扁杏仁全肽和扁杏仁谷蛋白肽对HUVECs均有显著的抑制作用(P<0.01),同时显著地减弱去甲肾上腺素对细胞增殖的促进作用(P<0.01);在不同剂量扁杏仁全肽和扁杏仁谷蛋白肽干预下培养细胞48h,与对照组相比,培养液中NO含量显著升高、ET含量显著降低(P<0.01),同时对去甲肾上腺素具有显著的拮抗作用(P<0.01)。结论扁杏仁全肽特别是扁杏仁谷蛋白肽对血管内皮功能具有很强的保护作用。
Objective To investigate the effect of almond peptide on the endothelial function of human umbilical vascular endothelial cells (HUVECs). Methods HUVECs were cultured in vitro. The proliferation of HUVECs was induced by different doses of Amygdalin and Amygdalin at doses of 150, 300 and 600 μg · mL -1, respectively. Nitric oxide (NO) and endothelium (ET) content of the test. Results Compared with the control group, all the doses of almond total peptide and almond gluten peptide inhibited the proliferation of HUVECs significantly (P <0.01), and significantly attenuated the effect of norepinephrine on cell proliferation (P <0.01). Compared with the control group, the content of NO in the culture medium was significantly increased and the content of ET was significantly decreased (P <0.01) after cultured with different doses of Amygdalin and Amygdalin for 48h, 0.01), while norepinephrine had a significant antagonistic effect (P <0.01). Conclusion Amygdalin, especially Amygdalin, has a strong protective effect on vascular endothelial function.
