目的对来自11个国家和中国2个省份不同时间分离的60株埃可病毒7型(ECHO virus 7,ECH-O 7)病毒VP1区基因特征进行分析。方法在GenBank中用BLASTN软件搜索不同国家、不同时间发表的ECHO 7VP1区基因序列,共搜索到97个序列,去除相同序列后,共得到60株ECHO 7病毒VP1区序列,用MEGA软件计算不同毒株的核苷酸和氨基酸差异率,并构建系统发生树。结果 60株病毒可分为4个基因型(基因型A~D),A基因型单独由ECHO 7原型株Wallace组成,B、C基因型各由1个亚型组成,D基因型由3个亚型组成,每个亚型又分为不同的组。不同毒株的核苷酸差异率为5.9%~23.2%(氨基酸差异率为1.0%~13.0%),不同基因型之间的核苷酸差异率为15.4%~22.5%(氨基酸差异率为1.7%~7.3%)。结论 ECHO 7病毒存在不同的基因型/亚型,不同基因型/亚型可以在同一个地区流行很长时间,同一时间内不同的基因型/亚型又可在不同的地区流行,具有明显的时间和地域流行特点。
Objective To analyze the characteristics of VP1 gene of 60 ECHO virus 7 (ECHO7) viruses isolated from 11 countries and 2 Chinese provinces at different times. Methods In GenBank, BLASTN software was used to search the ECHO 7VP1 gene sequences published in different countries at different times. A total of 97 sequences were searched out. After removing the same sequences, a total of 60 ECHO 7 VP1 region sequences were obtained. Using MEGA software, Strain nucleotide and amino acid difference rates, and the construction of phylogenetic tree. Results The 60 isolates were divided into 4 genotypes (genotypes A to D). A genotype was composed of ECHO 7 prototype Wallace, B and C genotypes consisted of 1 subtype, and D genotype consisted of 3 Subtypes, each subtype is divided into different groups. The nucleotide variation rates of different strains ranged from 5.9% to 23.2% (amino acid difference rate was 1.0% ~ 13.0%). The nucleotide differences among different genotypes ranged from 15.4% to 22.5% (amino acid difference rate was 1.7 % ~ 7.3%). Conclusion There are different genotypes / subtypes of ECHO 7 virus. Different genotypes / subtypes may prevail in the same area for a long time. Different genotypes / subtypes may be endemic in different areas at the same time, with obvious Time and geographical features.
