目的采用HPLC法测定复方磷酸可待因糖浆中的有关物质吗啡和防腐剂。方法采用Symmetry C18色谱柱(250 mm×4.6 mm,5μm),以含2.5 mmol·L-1庚烷磺酸钠的0.01 mol·L-1磷酸二氢钾溶液(含0.1%三乙胺,磷酸调p H2.5)-乙腈(80∶20)为流动相,检测波长210 nm,柱温30℃,进样量20μL。结果吗啡、苯甲酸在测定范围内的线性关系良好(r>0.9997),平均回收率分别为98.6%、100.9%(n=9)。结论所用方法简单、可靠,适用性好,可用于复方磷酸可待因糖浆的质量控制。
Objective To determine the related substances morphine and preservative in compound codeine phosphate syrup by HPLC. Methods A Symmetry C18 column (250 mm × 4.6 mm, 5 μm) was used with 0.01 mol·L -1 potassium dihydrogen phosphate solution containing 2.5 mmol·L -1 heptane sulfonate (containing 0.1% triethylamine and phosphoric acid Tune pH2.5) - acetonitrile (80:20) as the mobile phase, the detection wavelength was 210 nm, the column temperature was 30 ℃ and the injection volume was 20μL. Results The linear relationship between morphine and benzoic acid was good (r> 0.9997). The average recoveries were 98.6% and 100.9% (n = 9), respectively. Conclusion The method is simple, reliable, and has good applicability. It can be used for quality control of codeine phosphate compound.
