香椿子总多酚对佐剂型关节炎大鼠的治疗作用

目的研究香椿子总多酚(TSTP)对佐剂型关节炎大鼠的治疗作用。方法 6周龄Wistar大鼠50只,SPF级,随机分为5组,每组10只:正常组、模型组、雷公藤甲素组、TSTP低剂量组和TSTP高剂量组。正常组在左后肢足垫部皮下注射0.1 mL生理盐水,其余组在左后肢足垫部皮下注射0.1 mL弗氏完全佐剂(Freund’s complete adjuvant,FCA)诱导建立大鼠佐剂型关节炎模型。大鼠致炎后第8天开始灌胃治疗,正常组和模型组均给予0.5%羧甲基纤维素钠(10 mg·kg-1)、雷公藤甲素组给予雷公藤甲素(20 mg·kg-1)、TSTP低剂量组给予TSTP(35 mg·kg-1)、TSTP高剂量组给予TSTP(70 mg·kg-1),1次·d1,连续灌胃21 d。大鼠于致炎前(d0)、致炎后第4,8,12,16,20,24,28天(d4,d8,d12,d16,d20,d24,d28)分别在足容积测量仪上测定右侧(非致炎侧)足容积,d4,d8,d12,d16,d20,d24,d28与d0足容积的差值即为该时大鼠足趾肿胀度。最后一次灌胃结束2 h后,处理大鼠,同时摘取胸腺和脾脏组织并称重,计算胸腺指数和脾脏指数。并于踝关节上方0.5 cm剪取踝关节,采用苏木精-伊红染色法制作病理组织切片,观察形态学的相关改变。结果与模型组比较,TSTP降低了脏器指数、足跖肿胀度的水平,差异有统计学意义(P<0.05)。与雷公藤组相比,TSTP高剂量组脏器指数、足跖肿胀度无明显差别;与TSTP高剂量组相比,TSTP低剂量组脏器指数、足跖肿胀度较高(P<0.05)。结论 TSTP高,低剂量各组均能够减轻佐剂关节炎大鼠的足趾肿胀度、降低脾脏指数和胸腺指数,改善踝关节病理组织形态。TSTP对佐剂型关节炎大鼠具有一定的治疗作用。 Objective To study the therapeutic effect of TSTP on adjuvant arthritis in rats. Methods Fifty 6-week-old Wistar rats were randomly divided into 5 groups with 10 rats in each group: normal group, model group, triptolide group, low dose TSTP group and high dose TSTP group. In the normal group, 0.1 mL saline was injected subcutaneously into the footpad of the left hindlimb. The rest rats were injected subcutaneously with 0.1 mL Freund’s complete adjuvant (FCA) in the left hind paw footpad to induce the adjuvant arthritis model in rats. The rats were intragastrically administrated on the 8th day after inflammation. The rats in the normal group and model group were given 0.5% sodium carboxymethyl cellulose (10 mg · kg-1), and the triptolide triptolide (20 mg · kg- · Kg-1). TSTP (35 mg · kg-1) was given to low dose TSTP group and high dose TSTP group was given TSTP (70 mg · kg-1) once a day for 21 days. Rats were pre-inflated (d0), 4,8,12,16,20,24,28 days after inflammation (d4, d8, d12, d16, d20, d24, d28) Determination of right (non-inflammatory side) foot volume, d4, d8, d12, d16, d20, d24, d28 and d0 foot volume difference is the degree of rat toe swelling. Two hours after the last gavage, the rats were treated, the thymus and spleen tissues were harvested and weighed, and the thymus index and spleen index were calculated. Ankle joints were cut 0.5 cm above the ankle. Histopathological sections were made by hematoxylin-eosin staining and morphological changes were observed. Results Compared with the model group, TSTP reduced the level of organ index and paw edema, the difference was statistically significant (P <0.05). Compared with Tripterygium wilfordii group, there was no significant difference in organ index and paw edema between high dose TSTP group and high dose TSTP group (P <0.05) . Conclusion High and low doses of TSTP can alleviate the degree of the toe swelling, reduce the spleen index and thymus index of the adjuvant arthritis rats, and improve the morphology of the ankle joint. TSTP on adjuvant arthritis rats have a certain therapeutic effect.
