背景与目的CD44基因蛋白是一种分布极为广泛的细胞表面跨膜糖蛋白,它的变异多样性表达与肿瘤的生长及转移密切相关。本研究的目的是探讨CD44的表达、DNA含量(DI)、增殖指数(PI)和细胞凋亡率在女性肺腺癌中的关系。方法应用流式细胞仪(FCM)测定61例女性肺腺癌及其癌旁肺组织和45例肺部良性病变组织中CD44的表达、DI、PI和细胞凋亡率。结果女性肺腺癌组DNA异倍体出现率为75.41%。肺癌组中CD44的表达、DI值和PI值明显高于癌旁对照组和良性对照组(P<0.01),肺癌组细胞凋亡率则显著低于癌旁对照组和良性对照组(P<0.01)。肺癌组中CD44的表达与DI值呈正相关(P<0.01),凋亡率与DI值呈负相关(P<0.01)。结论CD44表达、DNA含量、增殖活性和细胞凋亡等因素在女性肺腺癌的发生、发展和转移过程中具有重要的作用。
BACKGROUND & OBJECTIVE: CD44 gene protein is a widely distributed cell surface transmembrane glycoprotein whose diversity of expression is closely related to tumor growth and metastasis. The purpose of this study was to investigate the relationship between CD44 expression, DNA content (DI), proliferation index (PI) and apoptosis in female lung adenocarcinoma. Methods Flow cytometry (FCM) was used to determine the expression of CD44, DI, PI and apoptosis in 61 cases of lung adenocarcinoma and its adjacent tissues and 45 cases of benign lung lesions. Results The frequency of DNA aneuploidy in female lung adenocarcinoma was 75.41%. The expression of CD44, DI and PI in lung cancer group were significantly higher than those in adjacent non-cancerous and benign control groups (P <0.01). The apoptosis rate in lung cancer group was significantly lower than that in adjacent non-cancerous and benign control groups (P < 0.01). There was a positive correlation between the expression of CD44 and DI in lung cancer group (P <0.01), and the negative correlation between apoptosis rate and DI value (P <0.01). Conclusion The expression of CD44, DNA content, proliferative activity and apoptosis have an important role in the occurrence, development and metastasis of human lung adenocarcinoma.
