目的:探讨IL-12对肥大细胞蛋白酶激活受体(PARs)表达的影响。方法:不同浓度IL-12刺激肥大细胞后,在不同时间点,用实时定量逆转录聚合酶链式反应(q-RT-PCR)和流式细胞术(FCM),在mRNA水平和蛋白水平检测PAR-1、PAR-2、PAR-3和PAR-4在肥大细胞P815表面和胞内的表达情况。结果:IL-12下调肥大细胞膜表面PAR-2蛋白的表达,上调肥大细胞膜表面和胞质内PAR-4蛋白的表达,IL-12抗体的应用可阻断IL-12对肥大细胞PARs蛋白表达的影响;IL-12上调肥大细胞PAR-1、3、4 mRNA的表达,下调PAR-2 mRNA的表达。结论:IL-12在过敏性炎症反应中的调节作用可能与IL-12调节肥大细胞PARs表达有关。
Objective: To investigate the effect of IL-12 on the expression of mast cell protease-activated receptors (PARs). Methods: After stimulation of mast cells with different concentrations of IL-12, mRNA and protein levels were detected by q-RT-PCR and flow cytometry (FCM) at different time points PAR-1, PAR-2, PAR-3 and PAR-4 on the surface and intracellular of mast cells P815. Results: IL-12 down-regulated the expression of PAR-2 protein on the surface of mast cell membrane and up-regulated the expression of PAR-4 protein on the surface and cytoplasm of mast cells. IL-12 antibody could block the expression of PARs protein in mast cells IL-12 up-regulated the expression of PAR-1,3,4 mRNA and down-regulated the expression of PAR-2 mRNA in mast cells. CONCLUSION: The regulatory role of IL-12 in allergic inflammation may be related to the regulation of PARs expression in mast cells by IL-12.
