目的:观察安胰颗粒对重症胰腺炎大鼠NF-κB活化的影响。方法:将120只大鼠分为正常组、模型组、安胰颗粒组和阳性对照组(IL-10组)共4组,各组又分为3 h、6 h、12 h三个亚组。采用6%的左旋精氨酸(L-Arginine)溶液诱导SAP大鼠模型。安胰颗粒组予安胰颗粒4 g/kg连续3天灌胃后诱导SAP。IL-10组予腹腔注射10000U人重组白介素10(rh IL-10)预处理后诱导SAP。摘取胰腺组织观察组织形态学变化并通过免疫组化检测胰腺组织NF-κBp65的表达。结果:安胰颗粒组及IL-10组病理学评分较模型组明显下降(P<0.05)。模型组胰腺组织NF-κBp65各时点表达显著高于正常组(P<0.01);3h即有NF-κBp65表达,呈现时间依赖性,6 h与3 h、12 h时间点比较,差异有统计学意义(P<0.05);安胰颗粒组NF-κBp65各时点表达较模型组明显下降(P<0.05),12 h时点表达较IL-10组明显下降(P<0.05)。结论:安胰颗粒能有效改善重症胰腺炎大鼠胰腺损伤,抑制NF-κB的表达可能是其作用机制之一。
Objective: To observe the effect of AnYi granule on activation of NF-κB in severe pancreatitis rats. Methods: A total of 120 rats were divided into 4 groups: normal group, model group, pancreas group and positive control group (IL-10 group), and each group was divided into 3 subgroups: 3 h, 6 h and 12 h . SAP rat model was induced by 6% L-arginine solution. Anan granule group to An Pan Granules 4 g / kg for 3 days after instilled gastric SAP. The IL-10 group was intraperitoneally injected with 10000 U of human recombinant interleukin 10 (rh IL-10) pretreatment induced SAP. Pancreatic tissues were removed for morphological changes and the expression of NF-κBp65 in pancreatic tissues was detected by immunohistochemistry. Results: Compared with the model group, the pathological score of An Pan Granules group and IL-10 group decreased significantly (P <0.05). The expression of NF-κBp65 in pancreatic tissue of model group was significantly higher than that of normal group (P <0.01). The expression of NF-κBp65 in 3h was in a time-dependent manner. There was statistical difference between 6h and 3h and 12h (P <0.05). The expression of NF-κBp65 at various time points in the Amniotic Granules group was significantly lower than that in the model group (P <0.05), and was significantly decreased at 12 h compared with the IL-10 group (P <0.05). Conclusion: AnYi granule can effectively improve pancreatic injury and inhibit the expression of NF-κB in severe pancreatitis rats.
