目的研究刺五加多糖(Acanthopanax senticosus polysaccharide,ASPS)对大鼠海马细胞的抗凋亡作用,并探讨其机制。方法运用海马神经细胞原代培养技术,采用H2O2诱导建立细胞凋亡模型。观察细胞形态学变化;MTT法测定细胞活性;免疫组化法检测细胞中Bax与Bcl-2的蛋白表达。结果形态学观察显示:与模型组比较,ASPS组细胞损伤程度明显减轻;ASPS组细胞活性(0.46)比模型组细胞活性(0.36)高(P<0.01),ASPS组Bcl-2表达量较高而Bax表达量较低。结论ASPS能够提高海马神经细胞的抗凋亡能力,并具有一定的浓度依赖性。
Objective To study the anti-apoptosis effect of Acanthopanax senticosus polysaccharide (ASPS) on rat hippocampal cells and to explore its mechanism. Methods Hippocampal neural cells were used for primary culture and H2O2 was used to induce apoptosis. Morphological changes of the cells were observed; cell viability was measured by MTT assay; protein expression of Bax and Bcl-2 was detected by immunohistochemistry. Results Morphological observations showed that compared with the model group, the degree of cell damage was significantly reduced in the ASPS group; the cell activity in the ASPS group (0.46) was higher than that in the model group (0.36) (P<0.01), and the Bcl-2 expression was higher in the ASPS group. Bax expression is low. Conclusion ASPS can increase the anti-apoptosis ability of hippocampal neurons and has certain concentration dependence.
