目的 比较心房钠尿肽 (ANP)、血管钠肽 (VNP)及C-型钠尿肽 (CNP)抑制心肌细胞增殖的效应 .方法 体外培养 SD乳鼠心肌细胞 ,用蛋白激酶 C激动剂佛波酯 (PMA)刺激心肌细胞增殖 ,以总蛋白含量和噻唑蓝 (MTT)比色吸光度值为指标 ,观察 3种钠尿肽抑制心肌细胞增殖的效应 .结果 PMA(10 - 9~ 10 - 7mol· L- 1 )致使心肌细胞的总蛋白含量和MTT吸光度值显著升高 (P<0 .0 5 ) ,ANP(10 - 8~ 10 - 6 mol·L- 1 ) ,VNP和 CNP(10 - 7~ 10 - 6 mol· L- 1 )均可显著降低PMA(10 - 8mol· L- 1 )刺激的心肌细胞总蛋白含量和 MTT吸光度值 (P<0 .0 5 ) .结论 PMA对心肌细胞有促增殖作用 ,ANP,VNP和 CNP均可抑制 PMA刺激的心肌细胞增殖 ,抑制效应以 ANP最强
Objective To compare the effects of atrial natriuretic peptide (ANP), vascular natriuretic peptide (VNP) and C-type natriuretic peptide (CNP) on the proliferation of cardiomyocytes in vitro.Methods Cardiomyocytes from SD neonatal rats were cultured in vitro and treated with the protein kinase C agonist (PMA) to stimulate the proliferation of cardiomyocytes, the effects of three kinds of natriuretic peptide on the inhibition of cardiomyocyte proliferation were observed by the total protein content and MTT colorimetric absorbance.Results PMA (10 -9 ~ 10 -7 mol · (P <0.05), ANP (10 - 8 ~ 10 - 6 mol·L - 1), VNP and CNP (10 - 7) ~ 10 -6 mol·L -1) could significantly decrease the total protein content and the MTT absorbance (P <0.05) of myocardial cells stimulated by PMA (10 -8 mol·L -1) Proliferation, ANP, VNP and CNP all inhibited PMA-stimulated cardiomyocyte proliferation with the strongest inhibitory effect on ANP
