目的 观察不同程度烫伤大鼠血浆中皮质酮的水平及α -MSH和合成肽KPV对它的调节作用。方法 采用放射免疫法检测正常对照组、轻度烫伤组和重度烫伤组大鼠血浆皮质酮的浓度 ;采用体内放射TNF -α、IL - 1β中和抗体和α -促黑色素细胞刺激激素 (α-melanocyte-stimulatinghormone ,α -MSH)和合成肽KPV(Ac -D -Lys -L -Pro -D -Val)等措施调节其改变。结果 与正常对照组的血浆皮质酮水平 (15 4 32± 5 9 12ng/mL)相比 ,烫伤后 12h轻烫组的血浆皮质酮浓度 (134 4± 376 47ng/mL)显著升高 (P <0 0 1) ;与轻烫组相比 ,重烫组大鼠血浆皮质酮水平 (2 6 80± 44 3 2 3ng/mL)升高更加明显 (P <0 0 1)。体内注射TNFα、IL - 1β中和抗体和α-MSH及KPV均能明显降低重烫组动物血浆皮质酮水平。结论 重度烫伤可使大鼠血浆出现高皮质酮血症 ,体内注射TNFα、IL - 1β中和抗体及α -MSH和KPV均可在一定程度上减轻重度烫伤所致的高皮质酮血症
Objective To observe the levels of corticosterone in plasma of scalded rats and the regulatory effect of α-MSH and synthetic peptide KPV on it. Methods The levels of plasma corticosterone in normal control group, mild scald group and severe scald group were determined by radioimmunoassay. Neonatal human TNF - α, IL - 1β neutralizing antibody and α - melanocyte stimulating hormone (α- melanocyte-stimulatinghormone, α-MSH) and synthetic peptide KPV (Ac-D -Lys -L -Pro -D -Val). Results Plasma corticosterone concentrations (134 4 ± 376 47 ng / mL) in the scald group were significantly higher than those in the normal control group (15 4 32 ± 5 9 12 ng / mL) Compared with light scalded group, plasma corticosterone level increased more significantly (P <0.01) in heavy-heat group than that in light-scalded group. In vivo injection of TNFα, IL - 1β neutralizing antibodies and α - MSH and KPV significantly reduced plasma corticosterone levels in the scalded group. Conclusions Severe scalds can induce hypercortisolism in plasma of rats. In vivo injection of TNFα, IL - 1β neutralizing antibodies and α-MSH and KPV can both relieve hypercortisonemia due to severe burns to a certain extent
