目的用DOI诱导大鼠头部抽动建立Tourette综合征动物模型,研究该动物模型脑内VMAT2的表PP达。方法 30只Wistar大鼠随机分为对照组、连续腹腔注射DOI14d和21d组,应用RT-PCR法检测黑质VMAT2mRNA表达,应用免疫组织化学法观察大鼠纹状体VMAT2蛋白的表达。结果大鼠注射DOI14d和21d后黑质内VMAT2mRNA和蛋白表达水平与正常对照组显著增加(P<0.05),但14d和21d组之间比较无显著差异(P>0.05)。结论 DOI诱导的大鼠头部抽动模型脑内VMAT2过表达。
Objective To establish animal model of Tourette’s syndrome by DOI-induced head twitch in rats and study the expression of VMAT2 in the brain of this animal model. Methods Thirty Wistar rats were randomly divided into control group, continuous intraperitoneal injection of DOI14d and 21d group. The expression of VMAT2 mRNA in substantia nigra was detected by RT-PCR. The expression of VMAT2 protein in rat striatum was observed by immunohistochemistry. Results VMAT2 mRNA and protein expression in substantia nigra was significantly increased after DOI 14d and 21d (P <0.05), but there was no significant difference between the 14d and 21d groups (P> 0.05). Conclusion DOI induced VMAT2 overexpression in rat brain twitching model.
