三色堇花色艳丽,品种丰富,是重要的冬春季花卉。本研究采用黄底紫斑品种‘梦蝶’为研究材料,克隆花瓣中可能参与花青素累积起调控作用的MYB基因VwMYB8,结果显示:克隆得到的cDNA全长为1 078 bp,经ORF Finder分析发现其有长度为849 bp的开放阅读框,编码282个氨基酸;构建瞬时表达载体pXCS-VwMYB8,采用粒子轰击的方法作用于月季‘粉扇’花瓣细胞,经过48 h培养后观察转化材料细胞中色素的变化,转化的细胞颜色明显加深,表明基因VwMYB8可以提高月季花瓣细胞中色素的含量。
Pansy beautiful flowers, rich variety, is an important spring and winter flowers. In this study, yellowish purple species “Mengdie” was used as the material to clone the MYB gene VwMYB8 which may play a role in the regulation of anthocyanin accumulation.
