[目的]建立人体各种正常组织的流式细胞术细胞动力学参数。[方法]用流式细胞术检测337例恶性肿瘤或良性疾病患者某些正常组织的DNA指数(DI)、DNA倍体类型、细胞凋亡水平和增殖活性。[结果]在337例患者的正常组织中DI值为1.00±0.00,DNA倍体为2倍体 ,其细胞凋亡水平为2.67%±2.50% ,S期细胞比率为5.00%±4.59 %。在不同类组织中 ,某些组织的SPF检出率有显著性差异(P<0.05或P<0.01)。恶性肿瘤患者正常组织的Apo和SPF均显著高于良性疾病患者的正常组织(P<0.01和P<0.05)。[结论]在肿瘤研究中选择正常对照组织时 ,恶性肿瘤患者的“正常”组织未必能真正代表机体的正常组织 ;而选择良性疾病患者的正常组织作为肿瘤研究的正常对照组织 ,这种组织可能更接近无任何疾病的正常机体组织的生物学特性。
[Objective] To establish the flow cytometry kinetic parameters of human normal tissues. [Methods] The DNA index (DI), DNA ploidy, apoptosis and proliferative activity of some normal tissues in 337 patients with malignant tumors or benign diseases were detected by flow cytometry. [Results] The DI value of normal tissue in 337 patients was 1.00 ± 0.00, the DNA ploidy was 2-fold, the apoptosis level was 2.67% ± 2.50%, the S-phase cell ratio was 5.00% ± 4.59%. In different kinds of organizations, the detection rate of SPF in some organizations was significantly different (P <0.05 or P <0.01). The Apo and SPF of normal tissues of patients with malignant tumors were significantly higher than those of normal tissues (P <0.01 and P <0.05). [Conclusion] The “normal” tissue of patients with malignant tumors may not truly represent the body’s normal tissues when selecting normal control tissues in cancer research. The normal tissues of patients with benign diseases as the normal control tissue for tumor research may be Closer to the biological characteristics of normal body tissue without any disease.
