目的:测定线粒体融合蛋白2(Mitofusin 2,Mfn2)在正常早孕妇女和不明原因胚胎停止发育患者绒毛组织中的表达差异,探讨Mfn2在胚胎发育过程中的可能作用。方法:采用HE染色观察正常早孕妇女(正常早孕组,30例)和不明原因胚胎停止发育患者(胚胎停育组,35例)绒毛组织形态;采用免疫组织化学技术(SABC法)检测Mfn2在两组妇女绒毛组织中的定位表达和表达强度;同时采用实时荧光定量RT-PCR(qRT-PCR)技术检测两组妇女绒毛组织中Mfn2的相对表达。结果:Mfn2在正常早孕组和胚胎停育组绒毛合体滋养细胞和细胞滋养细胞中均有阳性表达,在细胞滋养细胞中表达尤为明显,而在间质细胞中无明显表达,且Mfn2阳性表达主要分布在细胞浆中。胚胎停育组的绒毛组织中Mfn2的相对表达量(0.006 8±0.003 2)显著低于正常早孕组(0.023 9±0.005 5),差异有统计学意义(P<0.01)。结论:Mfn2在不明原因胚胎停止发育患者绒毛组织中的表达水平下降,提示Mfn2可能与胚胎发育相关,绒毛滋养层细胞中Mfn2的正常表达可能对维持早期妊娠起到一定作用。
OBJECTIVE: To determine the difference of mitochondrial fusion protein 2 (Mfn2) expression in normal early pregnant women and unexplained villus tissues and to explore the possible role of Mfn2 in embryonic development. Methods: The morphology of villi in normal pregnant women (normal pregnancy group, 30 cases) and unexplained embryo stop development (35 cases in embryo suspension group) were observed by HE staining. Immunohistochemistry (SABC method) was used to detect the expression of Mfn2 in two The expression of Mfn2 in the villi of the two groups were detected by real-time fluorescent quantitative RT-PCR (qRT-PCR). CONCLUSION: The decreased expression of Mfn2 in the villi of patients with unexplained embryo stop suggests that Mfn2 may be involved in embryonic development. The normal expression of Mfn2 in villus trophoblast may play an important role in the maintenance of early pregnancy.
