目的:检测肿瘤细胞系和我国肿瘤患者癌组织中天冬氨酰-(天冬酰胺酰)β-羟化酶(ASPH)的分布及表达谱。方法:采用RT-PCR法检测肿瘤细胞系中ASPH基因的转录水平;细胞免疫荧光和Western blot方法检测肿瘤细胞系中ASPH蛋白的表达水平;免疫组织化学方法检测人癌组织和正常组织中ASPH的分布与表达。结果:在肿瘤细胞系中AS-PH在转录和翻译水平均有不同程度表达,ASPH蛋白分布于细胞膜和胞质区域;在肿瘤患者组织中有ASPH的高表达,而人正常组织中不表达或表达量极低。结论:ASPH是一种有潜力的广谱性肿瘤标记分子,有可能用于肿瘤的早期检测。
Objective: To detect the distribution and expression profile of aspartyl - (asparaginyl) β - hydroxylase (ASPH) in tumor cell lines and cancer patients in China. Methods: The transcription level of ASPH gene in tumor cell lines was detected by RT-PCR. The expression of ASPH protein in tumor cell lines was detected by immunofluorescence and Western blot. The expression of ASPH in human cancer and normal tissues was detected by immunohistochemistry Distribution and expression. Results: AS-PH was expressed at different transcriptional and translational levels in tumor cell lines. ASPH protein was distributed in the plasma membrane and cytoplasmic region. In the tumor tissues, there was high expression of ASPH, but not in normal human tissues or The amount of expression is very low. Conclusion: ASPH is a potential broad-spectrum tumor marker molecule that may be used for early detection of tumors.
