采用随机扩增多态性DNA(RAPD)技术,从100个随机引物中筛选出扩增效果较好的20个引物,分析桂林市23个桂花品种的基因组多态性。结果表明,20个随机引物共检测到193个位点,其中多态位点为114个,占59.1%。根据扩增结果对这些品种进行聚类分析,并构建出了树状聚类图,可将这些品种划分为4个品种群,与传统分类学结果一致。此项研究表明,以基因型为基础分析桂花品种间的区别是可行的。因此,RAPD技术为解决桂林市的桂花品种分类问题提供了重要依据。
Random amplified polymorphic DNA (RAPD) technique was used to screen 20 primers with good amplification from 100 random primers to analyze the genomic polymorphism of 23 sweet-scented osmanthus cultivars in Guilin. The results showed that a total of 193 loci were detected by 20 random primers, of which 114 polymorphic loci accounted for 59.1%. Based on the results of the amplification, the cluster analysis of these cultivars was carried out and a tree-like clustering map was constructed. These cultivars can be divided into 4 populations, which are consistent with the results of traditional taxonomy. This study shows that it is feasible to analyze the differences between sweet-scented osmanthus cultivars based on genotypes. Therefore, RAPD technology provides an important basis for solving the classification of osmanthus species in Guilin.
