目的:采用DNA条形码技术和随机多态性(RAPD)分子标记技术对12个不同品种(系)的白芍种质资源进行遗传多样性研究。方法:采用CTAB法提取白芍叶片基因组DNA,利用ITS2引物和RAPD随机引物对样品进行PCR扩增,对于ITS2-PCR体系,测序后采用相似性搜索(BLAST法)进行鉴定分析;对于RAPD-PCR体系,根据条带数目,应用NTSYS2.10e软件采用UPGMA法进行聚类分析。结果:12个品种的ITS2序列的BLAST结果高度一致,未见碱基位点的变异。RAPD引物碱基序列共扩增出308条带,其中多态性条带有257条,多态百分率为83.4%,各品种(系)间的相似系数在0.558~0.847之间。结论:白芍的各品种间保存着丰富的遗传多样性。
OBJECTIVE: To study the genetic diversity of white peony germplasm from 12 different varieties (lines) using DNA barcoding and random amplified polymorphic DNA (RAPD) technique. Methods: Genomic DNA was extracted from the leaves of white peony root by CTAB method. ITS2 primers and RAPD random primers were used to amplify the samples. ITS2-PCR system was sequenced and identified by BLAST method. For RAPD-PCR System, according to the number of bands, using NTSYS2.10e software using UPGMA clustering analysis. Results: The BLAST results of ITS2 sequences of 12 cultivars were highly consistent and no variation of base sites was found. A total of 308 bands were amplified with the base sequence of RAPD primers, of which 257 bands were polymorphic, with a percentage of polymorphism of 83.4%. The similarity coefficients of all the cultivars ranged from 0.558 to 0.847. Conclusion: There are abundant genetic diversity among the varieties of Radix Paeoniae Alba.
