目的:研究刺参多糖(HS)对谷氨酸(Glu)所致大鼠嗜铬细胞瘤PC12细胞损伤的保护作用。方法:用谷氨酸造成PC12细胞损伤模型,通过细胞形态观察、MTT、LDH、MDA、SOD测定,观察刺参多糖的保护作用。结果:15mmol/L谷氨酸作用PC12细胞24h,出现明显损伤,培养上清液中LDH含量增加,细胞SOD含量降低,MDA含量升高;50、150、450μg/mL刺参多糖可有效改善谷氨酸损伤引起的PC12细胞形态改变,提高细胞存活率,减少乳酸脱氢酶渗漏,提高SOD含量,降低MDA值;结论:刺参多糖对谷氨酸所致的PC12细胞损伤有保护作用。
Objective: To study the protective effect of polysaccharide (HS) on damage of PC12 cells induced by glutamate (Glu) in rat pheochromocytoma. Methods: PC12 cell injury model was induced by glutamate, and the protective effect of polysaccharides from stichopus japonicus was observed by morphological observation, MTT, LDH, MDA and SOD. Results: After exposed to 15 mmol / L glutamate for 24 h, the PC12 cells were damaged obviously. The content of LDH in the culture supernatant was increased, the content of SOD in the cells was decreased and the content of MDA was increased. The polysaccharides of 50, 150 and 450 μg / mL The morphological changes of PC12 cells were induced by the injury of glutamate, the cell viability was increased, the leakage of lactate dehydrogenase was decreased, the content of SOD was increased and the MDA content was decreased. Conclusion: The polysaccharides of Apostichopus japonicus could protect the PC12 cells induced by glutamate.
