目的:研究补肾健脾方干预对肾虚-黄体抑制流产模型大鼠内分泌-免疫的影响。方法:SPF级SD大鼠随机分为3组,每组15只。A组为补肾健脾方干预组,用羟基脲加米非司酮加补肾健脾方水提液灌服;B组为肾虚-黄体抑制流产模型组,用羟基脲加米非司酮灌服;C组灌服生理盐水。酶联免疫法检测大鼠血清孕酮(P)和子宫蜕膜组织的细胞因子白细胞介素-2(IL-2)、白细胞介素-10(IL-10)、转化生长因子-β1(TGF-β1)的浓度。结果:A组、B组流产率均显著高于C组(P<0.01);A组流产率显著低于B组(P<0.05)。A组第12天P显著高于B组(P<0.01),A组显著低于C组(P<0.05);B组显著低于C组(P<0.01)。A、B组的IL-2显著高于C组(P<0.05,P<0.01),B组的IL-10显著低于C组(P<0.01),A组的IL-10显著高于B组(P<0.01)。Th1/Th2细胞因子浓度B组显著高于A、C组(P<0.01);Th1/Th3细胞因子B组显著高于A、C组(P<0.01),A组和C组比较有差异(P<0.05)。结论:补肾健脾方通过提升血清孕激素分泌,促进母胎界面妊娠保护性Th2、Th3型细胞因子的分泌,调节免疫失衡,诱导免疫耐受,从而发挥抗流产效应。
Objective: To study the effects of Bushen Jianpi Recipe (FSW) on endocrine-immune system in rats with kidney deficiency-corpus luteum suppression model. Methods: SPF SD rats were randomly divided into 3 groups with 15 rats in each group. Group A was Bushen Jianpi Fang intervention group, with hydroxyurea plus mifepristone plus Bushen Jianpi Fang aqueous extract; Group B was kidney deficiency - luteal suppression abortion model group with hydroxyurea plus mifepristone ; C group fed with saline. Serum progesterone (P) and uterine decidua were detected by enzyme-linked immunosorbent assay (ELISA) for cytokines such as interleukin-2, interleukin-10 and transforming growth factor- -β1). Results: The abortion rates in group A and group B were significantly higher than those in group C (P <0.01). The abortion rate in group A was significantly lower than that in group B (P <0.05). P of group A on day 12 was significantly higher than that of group B (P <0.01), group A was significantly lower than group C (P <0.05), group B was significantly lower than group C (P <0.01). IL-2 in group A and B was significantly higher than that in group C (P <0.05, P <0.01), IL-10 in group B was significantly lower than that in group C (P <0.01) Group (P <0.01). The concentration of Th1 / Th2 cytokines in group B was significantly higher than that in group A and C (P <0.01), while the level of Th1 / Th3 cytokines in group B was significantly higher than that in group A and C (P <0.01) P <0.05). Conclusion: Bushenjianpi Recipe can exert anti-abortion effect by promoting serum progesterone secretion, promoting the secretion of protective Th2 and Th3 type cytokines during pregnancy in maternal-fetal interface, regulating immune imbalance and inducing immune tolerance.
