目的:基于多巴胺(DA)神经元功能亢进理论,观察愈癫汤对精神分裂症模型大鼠额前脑组织抗氧化应激防御系统相关指标的影响。方法:将48只雄性成年SD大鼠随机分为空白组、模型组、对照组及治疗组,每组12只。其中模型组、对照组及治疗组应用地卓西平腹腔注射造模,空白组应用0.9%氯化钠溶液行相同方法腹腔注射,连续14d;于第15天进行干预性给药,对照组给予0.036mg/m L利培酮悬浊液灌胃,治疗组给予0.92g/m L愈癫汤水煎剂灌胃,空白组、模型组给予相应体积蒸馏水灌胃,连续14d;检测各组大鼠额前脑组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH)、过氧化氢酶(CAT)活性及DA的表达情况。结果:治疗组大鼠额前组织内SOD、GSH及CAT活性明显高于模型组(P<0.05),而DA的表达明显低于模型组(P<0.05)。结论:愈癫汤可通过促进脑组织中SOD、GSH及CAT活性,将超氧化物转化成无毒的水,从而抑制自由基的形成,阻断氧化应激反应,从而逆转DA神经元功能亢进引起的氧化应激反应。
OBJECTIVE: To observe the effect of Yuyientang Decoction on related indicators of antioxidant defense system in frontal brain tissue of schizophrenia rats based on the theory of dopamine (DA) hyperactivity. Methods: 48 male adult SD rats were randomly divided into blank group, model group, control group and treatment group, 12 in each group. The model group, the control group and the treatment group were injected with dizocilpine intraperitoneally. The blank group was injected intraperitoneally with 0.9% sodium chloride solution for 14 days continuously. Conclusion: Yuyient Decoction can inhibit the formation of free radicals, block the oxidative stress reaction and reverse the hyperactivity of DA neurons by promoting the activity of SOD, GSH and CAT in brain tissue and converting superoxide into non-toxic water Induced oxidative stress response.
