目的:探讨核转录因子κB(NF-κB)、巨噬细胞移动抑制因子(MIF)与育龄期妇女子宫内膜息肉(EPs)发病的关系及其在EPs中表达的相关性。方法:收集宫腔镜手术中42例子宫内膜息肉患者的子宫内膜息肉、息肉周围内膜组织及22例中隔子宫患者正常内膜组织,采用逆转录聚合酶链反应(RT-PCR)技术检测NF-κBp65及MIFmRNA的表达,免疫组织化学S-P法检测NF-κBp65及MIF蛋白的表达。结果:EPs中NF-κBp65及MIF mRNA的表达均高于同期息肉周围内膜及正常子宫内膜(P<0.05),与正常子宫内膜相比,息肉周围内膜中NF-κBp65及MIF mRNA的表达均增强(P<0.05)。NF-κBp65及MIF蛋白在3组中均有表达,均以息肉组中的表达最强,息肉周围内膜组织中的表达次之,3组间两两比较差异有统计学意义(P<0.05)。NF-κBp65及MIF mRNA在EPs中的表达呈正相关(r=0.694,P<0.05),NF-κBp65及MIF蛋白在EPs中的表达呈正相关(r=0.37,P<0.05)。结论:NF-κB和MIF可能与育龄期妇女EPs的发病机制有关,协同参与了EPs发生发展的病理变化。
Objective: To investigate the relationship between nuclear factor κB (NF-κB), macrophage migration inhibitory factor (MIF) and the pathogenesis of endometrial polyps (EPs) in women of childbearing age and its correlation with the expression of EPs. Methods: Forty-two patients with endometrial polyps in hysteroscopic surgery were enrolled in this study. Endometrial polyps, endometrial polyps, and normal endometrium in 22 cases of uterine septum were collected. Reverse transcription-polymerase chain reaction (RT-PCR) The expression of NF-κBp65 and MIF mRNA was detected by immunohistochemistry and the expression of NF-κBp65 and MIF protein were detected by immunohistochemical SP method. Results: The expressions of NF-κBp65 and MIF mRNA in EPs were significantly higher than those in peripheral endometrium and normal endometrium in the same period (P <0.05). Compared with normal endometrium, NF-κBp65 and MIF mRNA (P <0.05). The expression of NF-κBp65 and MIF protein in all three groups were the highest, the highest expression in polyps group and the second in the endometrial tissue of polyps. The difference between the three groups was statistically significant (P <0.05 ). The expressions of NF-κBp65 and MIF mRNA in EPs were positively correlated (r = 0.694, P <0.05). The expressions of NF-κBp65 and MIF in EPs were positively correlated (r = 0.37, P <0.05). CONCLUSION: NF-κB and MIF may be related to the pathogenesis of EPs in women of childbearing age and synergistically participate in the pathological changes of EPs.
