目的 研究辐射远后效应对红细胞膜阴离子转运蛋白 (band 3)基因表达的影响。方法 分离 10年期骨髓型急性放射病 (ARS)患者骨髓有核细胞 ,二步法液体培养红系 ,取新生红细胞涂片 ,做band 3mRNA原位杂交。抽提外周血有核细胞总RNA ,进行RT PCR ,以 β actin作内参照 ,半定量分析band 3mRNA表达丰度。结果 ARS新生红细胞中band 3mRNA原位杂交显色不均、淡染 ,定量分析显示 1 2患者band 3mRNA表达强度减弱。结论 辐射远后效应可能在转录水平影响红细胞膜阴离子转运蛋白基因表达 ,使转录产物减少 ,翻译产物功能下降即阴离子转运能力减弱 ,红细胞成熟过程延迟
Objective To study the effect of far-after radiation on the expression of band 3 gene in erythrocyte membrane. Methods Bone marrow nucleated cells were isolated from 10-year old patients with acute myeloid leukemia (ARS). Two-step liquid culture was performed on erythrocytes and smeared with erythrocytes to make band 3 mRNA in situ hybridization. Total RNA of peripheral blood mononuclear cells was extracted and RT PCR was performed. Β 3 actin was used as an internal reference to semi-quantitatively analyze the expression abundance of band 3 mRNA. Results In situ hybridization of band 3 mRNA in erythrocytes of ARS showed uneven staining. The light staining and quantitative analysis showed that the expression of band 3 mRNA in ARS was diminished. Conclusion The far-after radiation effect may affect the gene expression of erythrocyte membrane anion transporter protein at transcriptional level, decrease the transcripts, decrease the translational product function, weaken the anion transport ability and retard the red blood cell maturation process
