目的建立黄热疫苗病毒17D株的Vero细胞适应株。方法将2批鸡胚黄热疫苗病毒在Vero细胞上进行2次蚀斑筛选,并连续传代培养,然后对获得的4株Vero细胞适应株病毒YFV-Vero(5)-A1、YFV-Vero(5)-A2、YFV-Vero(5)-B1和YFV-Vero(5)-B2进行稳定性和免疫原性检测。结果4株Vero细胞适应株病毒可被黄热病毒(17D)猴免疫血清中和。在传代过程中,不同代次的病毒滴度稳定在6·90LgPFU/ml以上,高峰病变出现时间为5~7d。家兔和小鼠的免疫血清中和抗体效价分别为1∶640~1∶5120和1∶640~1∶2560。结论4株Vero细胞适应株病毒生物学性状稳定,免疫原性良好。
Objective To establish a Vero cell-adapted strain of yellow fever vaccine strain 17D. Methods Two batches of chick embryo yellow fever vaccine virus were screened twice on Vero cells and subcultured continuously. After that, four strains of Vero cell-adapted virus YFV-Vero (5) -A1 and YFV-Vero 5) -A2, YFV-Vero (5) -B1 and YFV-Vero (5) -B2 for stability and immunogenicity. Results Four strains of Vero cell adapted virus could be neutralized by yellow fever virus (17D) monkey immune sera. During passage, virus titers of different generations stabilized at above 6 · 90LgPFU / ml, and peak lesions occurred from 5 to 7 days. Rabbit and mouse immune serum neutralizing antibody titers were 1: 640 to 1: 5120 and 1: 640 to 1: 2560, respectively. Conclusion The four strains of Vero cells are stable in biological characteristics and have good immunogenicity.
