背景:白细胞介素17是一种强效的炎性细胞因子,在急性排斥反应研究中较多,而在慢性排斥反应以及急性和慢性共同研究甚少。目的:通过研究白细胞介素17表达与移植肾发生排斥反应的相关性,探索白细胞介素17表达与排斥反应的关系。方法:选取解放军第309医院肾移植后穿刺及复查患者47例,根据患者病情分为肾功能稳定组(n=20)、急性排斥组(n=15)和慢性排斥组(n=12),另取健康体检人员15人作为正常组。应用实时荧光定量PCR检测4组受试对象外周血单个核细胞中白细胞介素17 mR NA的相对表达水平,分析其与血清肌酐和排斥时间的相关性。结果与结论:与正常组相比,急性排斥组与稳定组患者白细胞介素17 m RNA的表达水平差异有显著性意义;而慢性排斥组患者白细胞介素17 mR NA较稳定组升高了2.89倍(P<0.01)。急性排斥组及慢性排斥组患者白细胞介素17 mR NA表达水平与肌酐、移植后排斥时间均无明显相关性。提示白细胞介素17在排斥反应中存在显著性意义。
BACKGROUND: Interleukin-17 is a potent, proinflammatory cytokine that is widely studied in acute rejection but rarely studied in chronic rejection and acute and chronic conditions. OBJECTIVE: To investigate the relationship between interleukin-17 (IL-17) expression and rejection in renal allograft and explore the relationship between interleukin-17 (IL-17) expression and rejection. Methods: Forty-seven patients with puncture and reexamination after renal transplantation were selected from the 309th Hospital of People’s Liberation Army. According to their condition, they were divided into stable renal function group (n = 20), acute rejection group (n = 15) and chronic rejection group Another health examination staff 15 people as a normal group. Real-time fluorescence quantitative PCR was used to detect the relative expression level of interleukin-17 mRNA in peripheral blood mononuclear cells of four groups, and its correlation with serum creatinine and rejection time was analyzed. RESULTS AND CONCLUSION: Compared with normal group, the expression level of IL-17 mRNA in acute rejection group and stable group was significantly different. In chronic rejection group, the IL-17 mR NA was 2.89 higher than that in stable group Times (P <0.01). The levels of IL-17 mRNA in acute rejection group and chronic rejection group had no significant correlation with creatinine and rejection time after transplantation. It is suggested that there is a significant significance of IL-17 in rejection.
